In postmenopausal women, there is substantial inter-individual variation in plasma estrogens. We have identified a variant SNP, associated with concentrations of estradiol, that creates a functional estrogen response element (ERE). GWAS in women receiving aromatase inhibitors for early breast cancer have identified variant SNPs, associated with musculoskeletal adverse events, that also create an ERE. GWAS in women receiving tamoxifen or raloxifene for breast cancer prevention revealed variant SNPs associated with risk and activity of BRCA1.
